T4 DNA Ligase catalyzes the formation of a phosphodiester bondbetween the terminal 5′ phosphate and 3′ hydroxyl groups of duplexDNA or RNA. The enzyme efficiently joins blunt and cohesive endsand repairs single stranded nicks in duplex DNA, RNA or DNA/RNAhybrids.

T4DNA连接酶催化双链DNA或RNA的末端5'磷酸和3'羟基之间的磷酸二酯键的形成。该酶有效地连接钝性和粘性末端并修复双链DNA，RNA或DNA/ RNA杂交体中的单链缺口。

The T4 DNA ligase is free of detectable of exo- and endonucleaseactivities, as judged by agarose gel electrophoresis followingincubation of 10 units of enzyme for 16 hours at 37 °C with 1 µg ofHindIII-digested λ DNA (Lanes 3 and 4) and supercoiled pUC19 DNA(Lanes 5 and 6).  Molecular weight markers are in Lanes 1 and2.
